Overview
Epitalon (Ala-Glu-Asp-Gly) is a synthetic tetrapeptide derived from the bovine pineal extract epithalamin, developed by Khavinson and colleagues at the St. Petersburg Institute of Bioregulation and Gerontology.
Telomerase Activation
In cultured human somatic cells, epitalon increased telomerase activity and produced measurable telomere lengthening over several passages — one of the few non-genetic, non-TERT-overexpression interventions reported to do so. The mechanism is proposed to involve direct binding to TERT promoter regions.
Pineal & Circadian Signalling
Epitalon restores age-associated declines in pineal melatonin secretion in rodent models, normalises circadian rhythm parameters and modulates GnRH pulsatility. The pineal-supportive mechanism is the basis for the original Khavinson hypothesis that pineal-derived signals coordinate age-related decline.
Gene Expression Effects
Microarray work reports epitalon modulates expression of genes involved in apoptosis (Bcl-2 family), antioxidant defence (SOD, catalase), and cell-cycle regulation (p53, p21), with directionality favouring cell survival under stress conditions.
Longevity Model Data
Multi-year rodent lifespan studies (Anisimov, Khavinson and colleagues) report 11–28% mean lifespan extension in mice across several genetic backgrounds. Spontaneous tumor incidence was reduced in several cohorts.

Cultured-cell work from the Khavinson group reports measurable telomere lengthening over multiple passages of human somatic cells. The finding has not been independently replicated to the same depth, and human clinical telomere-length data are limited.
How does epitalon support melatonin?
Rodent studies show epitalon restores age-associated declines in pineal melatonin secretion and normalises circadian parameters. The mechanism appears upstream of melatonin synthesis rather than direct melatonin receptor activity.
What does the longevity data show?
